FBXL20 also known as F-box and leucine-rich repeat protein 20 functions as part of the SCF (SKP1-cullin-F-box) ubiquitin ligase complex. This protein weighs approximately 45 kDa and plays an important role in protein degradation by facilitating ubiquitination marking proteins for proteasomal degradation. FBXL20 shows expression in various tissues including the brain and heart suggesting its role in multiple physiological processes.
The protein helps regulate important cellular processes such as cell cycle control and signal transduction. It acts as part of the larger SCF complex where it recognizes and binds specific phosphorylated substrates. Through these interactions FBXL20 maintains cellular homeostasis by targeting key regulatory proteins for degradation impacting overall cellular function.
FBXL20 integrates into the cell cycle and Wnt signaling pathways. In the cell cycle pathway it ensures proper progression through different phases by targeting cyclins for degradation. In the Wnt signaling pathway FBXL20 affects the stability of proteins like β-catenin important for proper signal transmission. It interacts with related proteins such as SKP1 and Cullin-1 within these pathways to execute its functions efficiently.
FBXL20 influences the development of cancer and neurodegenerative diseases. Altered FBXL20 function can disrupt regular protein degradation patterns leading to accumulations that contribute to tumorigenesis or neuronal damage. In cancer its interaction with proteins such as p53 can affect tumor suppression. Similarly in neurodegenerative diseases like Alzheimer's disrupted degradation pathways involving FBXL20 may play a role in disease progression.
