In Western blot, ab91650 was shown to bind specifically to FE65. A band was observed at 65/85 kDa in wild-type HEK-293T cell lysates with no signal observed at this size in APBB1 knockout cell line ab267294 (knockout cell lysate ab257833). To generate this image, wild-type and APBB1 knockout HEK-293T cell lysates were analysed. First, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 5 % milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed (ab216776) at 1/20000 dilution.

Biological function summary
FE65 plays a significant role in neuronal development and cognitive functions. It is part of a protein complex that includes the amyloid precursor protein (APP). This complex is important for the regulation of gene expression and cellular communication. FE65 also affects the nuclear signaling by interacting with the chromatin-remodeling complex. These interactions suggest that FE65 has a hand in influencing transcriptional activities within the cell.
Pathways
FE65 is involved in the APP processing pathway and the development of Alzheimer's disease. It interacts with APP and influences its role in the cleavage and formation of amyloid-beta peptides. These peptides are key factors within the Alzheimer's disease pathway. Moreover FE65 connects with other proteins such as Abl which is involved in cell differentiation and migration processes.
