Supplementary information

The FES protein also known as Feline Sarcoma Oncogene is a cytoplasmic tyrosine kinase with a molecular mass of approximately 93 kDa. It plays a role in regulating processes like cell growth differentiation and survival. FES is mainly expressed in hematopoietic cells endothelial cells and some neural tissues. Its structure includes SH2 and SH3 domains allowing the protein to interact with other cellular components.
Biological function summary

FES protein is involved in signal transduction pathways that control various physiological functions. It does not form part of any giant complexes rather it serves as a relay point for signals in cellular contexts. The kinase activity of FES is important for ensuring cellular processes such as actin remodeling an important process for cell movement and shape changes.

Pathways

The FES protein participates in the RAS and MAPK signaling pathways. These pathways are important for transmitting extracellular signals that govern cell fate decisions. FES interacts with proteins such as GRB2 and SOS1 in these pathways to modulate downstream effects and enable effective response to cellular stimuli.

Aberrant FES signaling has links to cancer and inflammatory disorders. Dysregulation of FES can contribute to several types of cancer including leukemia due to its effect on cell proliferation and survival. In inflammation FES interaction with proteins like STAT3 may promote pathological inflammatory responses therefore implicating FES in diseases such as rheumatoid arthritis.

Target data

Tyrosine-protein kinase that acts downstream of cell surface receptors and plays a role in the regulation of the actin cytoskeleton, microtubule assembly, cell attachment and cell spreading. Plays a role in FCER1 (high affinity immunoglobulin epsilon receptor)-mediated signaling in mast cells. Acts down-stream of the activated FCER1 receptor and the mast/stem cell growth factor receptor KIT. Plays a role in the regulation of mast cell degranulation. Plays a role in the regulation of cell differentiation and promotes neurite outgrowth in response to NGF signaling. Plays a role in cell scattering and cell migration in response to HGF-induced activation of EZR. Phosphorylates BCR and down-regulates BCR kinase activity. Phosphorylates HCLS1/HS1, PECAM1, STAT3 and TRIM28.
