FGF-15 also known as fibroblast growth factor 15 is a protein with a significant role in various physiological processes. This protein belongs to the fibroblast growth factor (FGF) family and has a molecular mass of approximately 25 kDa. FGF-15 is predominantly expressed in the intestines particularly in the ileum. Its main mechanical function involves the modulation of bile acid synthesis which occurs through its actions on specific receptor targets.
Biological function summary

FGF-15 plays a critical role in liver function and metabolism. It acts as a regulatory factor in bile acid homeostasis and glucose metabolism ensuring the balance and maintenance within the body. FGF-15 does not primarily function as part of a protein complex but it exerts its influence by binding to the fibroblast growth factor receptor 4 (FGFR4) associated with the β-Klotho co-receptor eliciting a signaling cascade important for maintaining metabolic homeostasis.

Pathways

FGF-15 engages in the enterohepatic circulation of bile acids and has a pivotal role in the suppression of bile acid synthesis. One of the key pathways involves fibroblast growth factor receptor signaling pathway. In this capacity FGF-15 works in conjunction with proteins such as FGFR4 and β-Klotho forming a signaling axis that influences the expression of cytochrome P450 family 7 subfamily A member 1 (CYP7A1) an important enzyme in bile acid biosynthesis.

FGF-15 is closely associated with metabolic conditions particularly liver diseases such as non-alcoholic fatty liver disease (NAFLD). Elevated levels of protein expression or dysregulation of its pathway may contribute to the development or progression of such disorders.
