FGF1 alpha also known as fibroblast growth factor 1 or aFGF functions as an important mediator in cellular processes such as proliferation differentiation and angiogenesis. This protein with a mass of approximately 18 kDa binds to specific fibroblast growth factor receptors (FGFRs) and heparan sulfate proteoglycans to initiate signal transduction. FGF1 alpha expresses in various tissues including brain liver and heart and plays significant roles in development and tissue repair.
Biological function summary

FGF1 alpha facilitates critical cellular functions by promoting cellular growth and survival. It often associates with other members of the fibroblast growth factor family and signaling molecules to form complexes essential for signal cascade initiation. This protein supports cellular resilience under stress conditions and participates in a variety of cellular activities which include wound healing and neovascularization.

Pathways

FGF1 alpha plays a significant role in the MAPK/ERK and PI3K/Akt signaling pathways. These pathways are fundamental for regulating cell cycle progression apoptosis and metabolic stability. FGF1 alpha interacts with proteins like FGF2 and FGFR1 which are involved in transmitting signals from the cell membrane to the nucleus impacting gene expression and cellular response.

FGF1 alpha is associated with cancer and metabolic disorders such as obesity-related insulin resistance. Its dysregulation or aberrant expression may result in uncontrolled cell growth typical of tumorigenic processes. FGF1 alpha often relates to FGF2 in oncogenic pathways where both proteins contribute to angiogenesis and tumor growth indicating their potential targets in therapeutic strategies for cancer treatment.

Plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Functions as a potent mitogen in vitro. Acts as a ligand for FGFR1 and integrins. Binds to FGFR1 in the presence of heparin leading to FGFR1 dimerization and activation via sequential autophosphorylation on tyrosine residues which act as docking sites for interacting proteins, leading to the activation of several signaling cascades. Binds to integrin ITGAV : ITGB3. Its binding to integrin, subsequent ternary complex formation with integrin and FGFR1, and the recruitment of PTPN11 to the complex are essential for FGF1 signaling. Induces the phosphorylation and activation of FGFR1, FRS2, MAPK3/ERK1, MAPK1/ERK2 and AKT1 (PubMed : 18441324, PubMed : 20422052). Can induce angiogenesis (PubMed : 23469107).
