Supplementary information

FGF12 also known as fibroblast growth factor 12 is a member of the fibroblast growth factor family and has a molecular mass of approximately 28 kDa. FGF12 is expressed in various human tissues notably in the brain and heart where it plays an essential role in cellular processes. Unlike many other FGF family members FGF12 does not have a heparin-binding domain. This distinction affects its interaction with local extracellular matrices and influences how it functions within cells.
Biological function summary

Fibroblast growth factors regulate cell differentiation growth and tissue repair. Though not a typical secreted growth factor FGF12 functions intracellularly where it contributes to neuronal signaling. FGF12 interacts with ion channels and proteins in the central nervous system suggesting its role in modifying electrical signaling pathways. It does not typically form part of a large complex but it associates with proteins that mediate such signaling functions.

Pathways

FGF12 plays a significant role in the intracellular signaling pathways especially those related to neurogenesis and cardiac conduction. In these pathways FGF12 regulates ion channel activity ensuring proper signal propagation. It connects with related pathways of other fibroblast growth factors such as FGF11 sharing key similarities in modulating electrophysiological properties within excitable cells like neurons and cardiomyocytes.

FGF12 has links to cardiac arrhythmias and epilepsy. Alterations in FGF12 expression can disrupt normal cardiac and neural electrical activity hinting at its involvement in these conditions. It interacts with several ion channels such as the sodium channel subunit SCN5A which can result in clinical manifestations when dysregulated. Understanding FGF12's role provides insights into potential therapeutic targets for managing arrhythmias and seizure disorders.
