FGFR2IIIb also known as FGFR2b is a variant of the fibroblast growth factor receptor 2 (FGFR2) with a molecular weight of approximately 120 kDa. This receptor is a critical component of the FGFR family and is expressed predominantly in epithelial tissues. It is one of the isoforms generated through alternative splicing distinguished by the inclusion of exon IIIb. FGFR2b plays a fundamental role in ligand binding with specific fibroblast growth factors leading to receptor dimerization and activation of its intrinsic tyrosine kinase activity.
Biological function summary

FGFR2b is important in the regulation of epithelial cell proliferation and differentiation. It participates in forming a receptor-ligand complex upon binding with its ligands which include FGF7 and FGF10. This receptor is involved in embryonic development organogenesis and tissue repair processes. FGFR2b influences cellular functions by activating downstream signaling cascades that dictate cell fate decisions. Its activity must be tightly regulated to ensure proper developmental and tissue-specific outcomes.

Pathways

FGFR2b is involved in the MAPK/ERK and PI3K/AKT signaling pathways two critical pathways associated with cell proliferation survival and differentiation. These pathways are activated upon FGFR2b dimerization and trans-autophosphorylation which recruits adaptor proteins like GRB2 and SOS1. FGFR2b shares common mechanistic connections with other growth factor receptors such as EGFR that cooperate in controlling cellular outcomes through these pathways. The FGFR2b signaling intersects with these pathways to ensure comprehensive cellular responses to environmental signals.

Defects or dysregulation in FGFR2b function are linked to various types of cancer including breast cancer and lung cancer. Abnormal FGFR2b signaling can lead to unchecked cell proliferation and tumor development. Additionally FGFR2b mutations are implicated in craniosynostosis syndromes like Apert syndrome where premature fusion of cranial sutures occurs. In these conditions FGFR2b is often found in conjunction with abnormal regulation of FGF ligands and interactions with proteins like FGF2. Targeting FGFR2b for therapeutic intervention is an active area of research in managing these disorders.

Tyrosine-protein kinase that acts as a cell-surface receptor for fibroblast growth factors and plays an essential role in the regulation of cell proliferation, differentiation, migration and apoptosis, and in the regulation of embryonic development. Required for normal embryonic patterning, trophoblast function, limb bud development, lung morphogenesis, osteogenesis and skin development. Plays an essential role in the regulation of osteoblast differentiation, proliferation and apoptosis, and is required for normal skeleton development. Promotes cell proliferation in keratinocytes and immature osteoblasts, but promotes apoptosis in differentiated osteoblasts. Phosphorylates PLCG1, FRS2 and PAK4. Ligand binding leads to the activation of several signaling cascades. Activation of PLCG1 leads to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5-trisphosphate. Phosphorylation of FRS2 triggers recruitment of GRB2, GAB1, PIK3R1 and SOS1, and mediates activation of RAS, MAPK1/ERK2, MAPK3/ERK1 and the MAP kinase signaling pathway, as well as of the AKT1 signaling pathway. FGFR2 signaling is down-regulated by ubiquitination, internalization and degradation. Mutations that lead to constitutive kinase activation or impair normal FGFR2 maturation, internalization and degradation lead to aberrant signaling. Over-expressed FGFR2 promotes activation of STAT1.
