FHL3 also known as Four and a Half LIM Domains Protein 3 functions as a transcriptional cofactor. It weighs approximately 30 kDa. FHL3 is expressed in skeletal muscle cardiac muscle and to some degree in other tissues such as the spleen and lung. This protein consists of LIM domains known for mediating important protein-protein interactions facilitating its role in cellular activities.
Biological function summary

The target plays an important role in transcriptional regulation and muscle development. FHL3 serves as part of transcriptional complexes and has been shown to interact with muscle-specific transcription factors. It promotes the correct expression of genes involved in the growth and maintenance of muscle tissues. Through these interactions FHL3 influences the structural and functional integrity of muscle cells.

Pathways

FHL3 operates within the Wnt signaling and the insulin-like growth factor (IGF) pathways. In these pathways it interacts with several proteins such as transcription factors like MyoD and Myogenin which are critical for muscle differentiation and regeneration. These interactions help in transmitting signals that modulate gene expression patterns essential for muscle function and repair.

Mutations or aberrant expression of FHL3 associates with muscular dystrophy and certain heart conditions. In muscular dystrophy alterations in FHL3 affect muscle fiber integrity often through its interactions with other myotonic dystrophy proteins. In cardiac conditions FHL3 involvement impacts the heart muscle structure and function through its connection with proteins involved in cardiac remodeling and stress response such as the protein titin.
