Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of Human colon carcinoma tissue sections labeling Fibroblast activation protein, alpha with ab227703 at 1/100 dilution (1.76 μg/ml). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0, epitope retrieval solution 2) for 10 mins. Rabbit specific IHC polymer detection kit HRP/DAB (ab209101) was used as the secondary antibody. Hematoxylin was used as a counterstain. Positive staining on the stroma in the human colon carcinoma, performed on a Leica Biosystems BOND™ RX instrument.
The section was incubated with ab227703 for 10 mins at room temperature.

This data was developed using ab227703, the same antibody clone in a different buffer formulation.

Western blot : Rabbit Monoclonal[SP325] to Fibroblast activation protein, alpha ab227703 staining at 0.176 µg/mL, shown in green; Mouse anti GAPDH ab8245 loading control staining at 1/20000 dilution, shown in magenta. A band was observed at 90-100 kDa in Wild-type U-87 MG cell lysates with no signal observed at this size in FAP knockout U-87 MG cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3pc Milk in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW and Goat anti-Mouse 680RD at 1/20,000 dilution.

Cell surface glycoprotein serine protease that participates in extracellular matrix degradation and involved in many cellular processes including tissue remodeling, fibrosis, wound healing, inflammation and tumor growth. Both plasma membrane and soluble forms exhibit post-proline cleaving endopeptidase activity, with a marked preference for Ala/Ser-Gly-Pro-Ser/Asn/Ala consensus sequences, on substrate such as alpha-2-antiplasmin SERPINF2 and SPRY2 (PubMed : 14751930, PubMed : 16223769, PubMed : 16410248, PubMed : 16480718, PubMed : 17381073, PubMed : 18095711, PubMed : 21288888, PubMed : 24371721).
