Interleukin 33 (IL-33) also known as IL1F11 or NF-HEV is a cytokine belonging to the IL-1 family. It is a protein with a molecular weight of approximately 30 kDa. IL-33 is mainly expressed in endothelial cells epithelial cells and fibroblasts. It exists in a full-length form in the nucleus and releases into the extracellular environment upon cellular damage or necrosis indicating its role as an alarmin. The expression of IL-33 can be upregulated in cells under stress or damaged conditions.
Biological function summary

In its role as a cytokine involved in immune response activation IL-33 functions as an alarmin signaling tissue damage or infection. It does not operate as part of a complex; instead IL-33 binds to the ST2 receptor (also known as IL1RL1) on target cells. This binding triggers the activity of immune cells such as type 2 helper T cells mast cells and innate lymphoid cells. By interacting with these immune cells IL-33 contributes to the body's inflammatory and repair processes controlling the early stages of immune response activation.

Pathways

IL-33 has a significant role in the signaling cascade related to the ST2/IL-1 receptor pathway. This pathway is essential for modulating inflammation and immune responses. Additionally IL-33 links with the NF-kB signaling pathway often involved in inflammatory responses. In these pathways its interactions with proteins like MyD88 and IRAK proteins are important for intracellular signaling and the promotion of cytokine production helping sustain the immune response.

IL-33 is associated with the pathogenesis of conditions such as asthma and rheumatoid arthritis. In asthma IL-33 contributes to airway inflammation and hypersensitivity acting in concert with other cytokines like IL-4 and IL-5 to amplify the type 2 immune response. In rheumatoid arthritis IL-33 engages with the ST2 receptor in joint tissues promoting inflammation and joint degradation. Such roles highlight IL-33's potential as a therapeutic target in modulating excessive immune responses in these diseases.

Cytokine that binds to and signals through the IL1RL1/ST2 receptor which in turn activates NF-kappa-B and MAPK signaling pathways in target cells (PubMed : 16286016, PubMed : 19841166). Involved in the maturation of Th2 cells inducing the secretion of T-helper type 2-associated cytokines (PubMed : 17853410, PubMed : 18836528). Also involved in activation of mast cells, basophils, eosinophils and natural killer cells (PubMed : 17853410, PubMed : 18836528). Acts as an enhancer of polarization of alternatively activated macrophages (PubMed : 19841166). Acts as a chemoattractant for Th2 cells, and may function as an 'alarmin', that amplifies immune responses during tissue injury (PubMed : 17853410, PubMed : 18836528). Induces rapid UCP2-dependent mitochondrial rewiring that attenuates the generation of reactive oxygen species and preserves the integrity of Krebs cycle required for persistent production of itaconate and subsequent GATA3-dependent differentiation of inflammation-resolving alternatively activated macrophages (By similarity).. In quiescent endothelia the uncleaved form is constitutively and abundantly expressed, and acts as a chromatin-associated nuclear factor with transcriptional repressor properties, it may sequester nuclear NF-kappaB/RELA, lowering expression of its targets (PubMed : 21734074). This form is rapidely lost upon angiogenic or pro-inflammatory activation (PubMed : 18787100).
