Function

Cortical cytoskeleton protein found in a complex involved in maintaining cell polarity and epithelial integrity. Involved in the regulation of mitotic spindle orientation, proliferation, differentiation and tissue organization of neuroepithelial cells. Involved in axonogenesis through RAB10 activation thereby regulating vesicular membrane trafficking toward the axonal plasma membrane.

Supplementary info

Activity summary

LLGL1 also known as LLGL Scribble Cell Polarity Complex Component 1 weighs approximately 156 kDa. It is an essential protein that helps maintain cell polarity and regulates tight junctions. LLGL1 is located mainly in the cytoplasm but can also localize to the cell membrane. Expression of LLGL1 occurs in various tissues with high levels found in brain skin and muscle tissue. As part of a cell polarity complex LLGL1 interacts with other proteins to control cell organization and architecture.

Biological function summary

LLGL1 plays a fundamental role in maintaining epithelial cell polarity and asymmetric cell division. These functions are essential for correct tissue organization and cellular differentiation. LLGL1 forms a complex with Scribble and Dlg1 which is important for determining apico-basal cell polarity. This complex helps cells achieve and maintain their functional orientation and is critical for tissue formation.

Pathways

LLGL1 interacts with pathways related to cell polarity and signaling. The protein is involved in the Hippo signaling pathway which is essential for regulating organ size by controlling cell proliferation and apoptosis. In this pathway LLGL1 interacts with proteins like YAP and TAZ which are critical for normal cellular functioning and development. The Par3/Par6/aPKC complex is another important pathway related to LLGL1 where its function ties into cellular asymmetry and differentiation.

Associated diseases and disorders

LLGL1 is linked to cancer and neurodevelopmental disorders. Its role in maintaining cell polarity and tissue architecture is disrupted in various cancers particularly in epithelial tumors suggesting a tumor suppressor function. Abnormalities in its function may result in loss of cell polarity which can contribute to tumor progression. In neurodevelopmental contexts alterations in LLGL1 can affect brain development and are associated with certain neurodevelopmental disorders. LLGL1 through its connection with proteins like Dlg1 and Scribble contributes to maintaining cellular organization that can prevent disorder pathogenesis.
