Activity summary

FKBP25 also known as FK506 Binding Protein 3 (FKBP3) is a peptidyl-prolyl cis-trans isomerase with a molecular mass of approximately 25 kDa. This protein mainly binds immunosuppressive drugs like tacrolimus and plays an important role in protein folding by accelerating the cis-trans conversion of proline residues in polypeptide chains. FKBP25 is broadly expressed in human tissues with higher expression levels reported in the heart liver and skeletal muscle indicating its significance in various cellular functions.

Biological function summary

FKBP25 interacts with ribosomal subunits and histones linking it to both ribosome assembly and chromatin regulation processes. It contributes to the formation of complexes involved in maintaining DNA stability and managing gene expression. Through these associations FKBP25 actively participates in cellular signaling and programmed gene transcription therefore significantly affecting cellular proliferation and differentiation mechanisms.

Pathways

FKBP25 participates in the mTOR signaling pathway impacting cell growth and metabolism. Additionally FKBP25 is involved in the folding and assembly of newly synthesized proteins through its interaction with the Hsp90 chaperone pathway. Its interaction with the mTOR complex as well as with Hsp90 and other co-chaperones highlights its integration into critical cellular survival and stress response mechanisms.

Associated diseases and disorders

FKBP25 has been linked to certain cancers and neurological diseases. Its interactions with mTOR and Hsp90 relate it to oncogenesis as these pathways are often dysregulated in tumor cells. Additionally alterations in FKBP25 function or expression have been associated with neurodegenerative disorders due to its critical role in protein homeostasis a process related to proteins such as mTOR and Hsp90 involved in cellular stress responses and synaptic signaling.
