ab129097 staining FKBP52 in MCF-7 (human breast carcinoma) cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% PFA and permeabilized with 0.1% triton X-100. Samples were incubated with primary antibody at a dilution of 1/500. A goat anti rabbit IgG (Alexa Fluor® 488) (ab150077) was used as the secondary antibody at a dilution of 1/1000.
DAPI was used as a nuclear counterstain and the negative control was PBS only.

Western blot : Anti-FKBP52 antibody [EPR6618] ab129097 staining at 1/1000 dilution, shown in green; Mouse anti GAPDH ab8245 loading control staining at 1/20000 dilution, shown in magenta. A band was observed at 52 kDa in Wild-type HCT 116 cell lysates with no signal observed at this size in FKBP4 knockout HCT 116 cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 3pc Milk in TBS-0.1 % Tween 20 (TBS-T) before incubation with primary antibodies overnight at 4 C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW and Goat anti-Mouse 680RD at 1/20,000 dilution.

Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.

Biological function summary
FKBP52 functions as a co-chaperone in steroid receptor complexes. It partners with heat shock protein 90 (HSP90) to form a functional complex that modulates the activity of steroid hormone receptors. These receptors such as the androgen and glucocorticoid receptors rely on FKBP52 for proper regulation and transport within the cell. FKBP52 also plays a part in cellular signaling influencing processes like cell growth and differentiation through its interaction with these receptors.
Pathways
FKBP52 features prominently in steroid hormone signaling and stress response pathways. It is critical to the maturation and transport of steroid hormone receptors directly affecting their downstream gene regulation.
