FKBP6 also known as FK506 binding protein 6 is a peptidyl-prolyl cis-trans isomerase. FKBP6 weighs approximately 24 kDa and is part of a larger family of FK506 binding proteins. It regulates protein folding and trafficking within the cell assisting in various cellular processes. FKBP6 is primarily expressed in testicular tissue indicating a significant role in reproductive system functions.
Biological function summary

FKBP6 associates with components of the synaptonemal complex during meiosis important for proper chromosomal pairing and segregation. FKBP6 associates with the protein TDRD6 helping in spermatogenesis. It may also play a role in regulating immune function through its interactions with other proteins in the FKBP family although this role is less understood.

Pathways

FKBP6 participates notably in the meiotic recombination pathway ensuring stability and proper meiotic division. It interacts with proteins like SPO11 and MLH1 both key in DNA processing during meiosis. FKBP6 also may interact with pathways related to cellular stress response but these interactions need further exploration.

FKBP6 has known associations with male infertility particularly cases involving azoospermia where sperm production ceases. Various studies suggest that disruptions in FKBP6 can lead to abnormal chromosomal behavior during meiosis contributing to this condition. The protein also shows potential links to psychiatric disorders such as schizophrenia possibly through connections with FKBP5 a protein believed to affect stress response pathways.

Has an essential role in spermatogenesis (PubMed : 36150389). It is required to repress transposable elements and prevent their mobilization, which is essential for the germline integrity (By similarity). Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and govern the methylation and subsequent repression of transposons (By similarity). Acts as a co-chaperone via its interaction with HSP90 and is required for the piRNA amplification process, the secondary piRNA biogenesis (By similarity). May be required together with HSP90 in removal of 16 nucleotide ping-pong by-products from Piwi complexes, possibly facilitating turnover of Piwi complexes (By similarity).
