Supplementary information

The FKSG14/p33 protein also known as Fuzzy Onions Homolog 2 or FUZ is a component involved in cellular processes like protein trafficking and vesicle transport. FKSG14 has a molecular weight of approximately 33 kDa. You often find it expressed in various tissues including neuronal and epithelial. Functionally FKSG14 plays a role in maintaining the dynamic organization of the cell through its involvement in cytoplasmic transport routes.
Biological function summary

FKSG14 participates in pathways linked to ciliogenesis and cellular asymmetry. It operates as part of the intraflagellar transport (IFT) complex which is essential in the development of primary cilia. These cilia play key roles in cell signaling and development. Presence of FKSG14 within the IFT complex underlines its importance to processes like organ lateralization and embryonic patterning. Its lack or dysfunction can disrupt normal signaling pathways critical for cellular and developmental functions.

Pathways

FKSG14 influences both the Hedgehog and Wnt signaling pathways. These pathways are important for cell differentiation and cell fate determination. Within these pathways FKSG14 interacts with proteins like Smo (Smoothened) in the Hedgehog pathway and Dvl (Disheveled) in the Wnt pathway. These relationships highlight FKSG14's involvement in complex signaling networks illustrating its key position in maintaining cellular integrity and function.

FKSG14 has links to conditions like ciliopathies and neural tube defects. Ciliopathies are disorders resulting from defects in cilia structure or function and can include polycystic kidney disease and Bardet-Biedl syndrome. FKSG14's disruption may connect with these disorders through its interaction with other ciliary proteins like IFT74. It also plays roles in neural development suggesting its involvement in neural tube defects when FKSG14 or related proteins like SHH (Sonic Hedgehog) pathways are impaired leading to congenital anomalies.

Target data

Component of the CENPA-CAD (nucleosome distal) complex, a complex recruited to centromeres which is involved in assembly of kinetochore proteins, mitotic progression and chromosome segregation. May be involved in incorporation of newly synthesized CENPA into centromeres via its interaction with the CENPA-NAC complex. Acts in coordination with KNL1 to recruit the NDC80 complex to the outer kinetochore.
