Supplementary information

Flightless I also known as FLII is a multifunctional protein with a mass of approximately 131 kDa. This protein belongs to the gelsolin family and contains leucine-rich repeats (LRR) and a partially functional gelsolin domain. It is widely expressed in various tissues including muscle and skin. Functionally Flightless I interacts with actin filaments and plays a role in modulating the actin cytoskeleton. Through its actin-binding property Flightless I participates in cellular processes such as motility and structural maintenance.
Biological function summary

Flightless I protein influences several cell mechanisms by associating with signaling complexes. It functions as a part of cellular assemblies where it modulates actin dynamics and affects transcriptional activities by interacting with other cellular components. Additionally Flightless I contributes to cell growth survival and wound healing processes. The protein can also regulate certain gene expressions by acting as a transcriptional co-regulator highlighting its involvement in cellular communication.

Pathways

Flightless I has significant roles in cellular signaling and cytoskeletal networks. It is actively involved in the Wnt signaling pathway as well as the Toll-like receptor signaling pathway. These pathways are central to cell communication and immune response regulation. Flightless I operates alongside proteins such as Dishevelled and MyD88 integrating signals that influence cell behavior and immune reactions. By mediating these interactions Flightless I enhances the cellular response to environmental cues.

Flightless I is associated with aberrant conditions like cancer and muscular dystrophy. Its dysregulation has been linked to tumor progression and metastasis where it influences cell migration and invasion. Flightless I interacts with proteins such as beta-catenin and dystrophin in these contexts affecting cellular adhesion and stability. Understanding these interactions can provide insights into therapeutic targeting strategies for managing and treating associated pathologies.

Target data

Is a regulator of actin polymerization, required for proper myofibril organization and regulation of the length of sarcomeric thin filaments (By similarity). It also plays a role in the assembly of cardiomyocyte cell adhesion complexes (By similarity). Regulates cytoskeletal rearrangements involved in cytokinesis and cell migration, by inhibiting Rac1-dependent paxillin phosphorylation (By similarity). May play a role as coactivator in transcriptional activation by hormone-activated nuclear receptors (NR) and acts in cooperation with NCOA2 and CARM1 (PubMed : 14966289). Involved in estrogen hormone signaling.
