FLIP-L also known as FLICE-like inhibitory protein-long is an important regulator in apoptosis. It has a molecular mass of approximately 55 kDa. FLIP-L is expressed in various tissues especially in lymphoid tissues and the spleen. It contains two death effector domains (DEDs) that hinder the protease activity of initiator caspases important components driving cell death pathways. By inhibiting caspase-8 FLIP-L prevents the initiation of apoptosis making it an important player in the control of cell death.
Biological function summary

FLIP-L inhibits apoptosis by interacting with the death-inducing signaling complex (DISC). It competes with procaspase-8 for binding within this complex reducing the activation of caspase-8 and subsequent apoptotic signals. This inhibition plays an important role in the regulation of immune responses and cancer progression. Through its involvement in apoptosis and immune signaling FLIP-L modulates cellular survival especially in the immune cell lineage where its function prevents excessive cell death.

Pathways

FLIP-L operates within the extrinsic apoptosis pathway and exhibits strong interactions with components like FADD (Fas-associated protein with death domain). It influences the balance between cell survival and death interacting with important players like caspases and TNF receptor signaling pathways. By modulating these interactions FLIP-L determines whether a cell follows a path towards apoptosis or survival reflecting its importance in maintaining cellular homeostasis.

FLIP-L has connections to cancer and autoimmune conditions. In many cancers elevated FLIP-L levels correlate with resistance to apoptosis hindering effective cancer treatments. Furthermore FLIP-L impacts autoimmune diseases by altering immune cell apoptosis leading to unchecked immune activity. Proteins such as Bcl-2 and caspase-8 are often discussed in connection with FLIP-L when analyzing its role in disease. By affecting these proteins FLIP-L contributes to the persistence and progression of various disease states.

Apoptosis regulator protein which may function as a crucial link between cell survival and cell death pathways in mammalian cells (PubMed : 31046799). Acts as an inhibitor of TNFRSF6 mediated apoptosis. A proteolytic fragment (p43) is likely retained in the death-inducing signaling complex (DISC) thereby blocking further recruitment and processing of caspase-8 at the complex. Full length and shorter isoforms have been shown either to induce apoptosis or to reduce TNFRSF-triggered apoptosis. Lacks enzymatic (caspase) activity.
