Activity summary

FOLR3 also known as folate receptor gamma is a member of the folate receptor family responsible for binding and transporting folate across cell membranes. This protein has a molecular mass of about 32 kDa. FOLR3 is primarily found in certain tissues including the placenta and liver and it exhibits limited expression in several epithelial tissues. Folate receptors including FOLR3 function by binding folates and delivering them into cells via endocytosis a process essential for the maintenance of regular cellular activity and function.

Biological function summary

Folate receptor gamma plays a significant role in the transport and uptake of folate an important vitamin necessary for DNA synthesis and methylation. The receptor assists in various cellular processes including nucleotide biosynthesis. FOLR3 works as part of a larger complex of folate receptors which ensure cells have adequate folate levels for normal metabolic functions. By doing so it helps facilitate normal cell growth and division critical in rapidly dividing cells.

Pathways

FOLR3 is notably involved in the folate metabolism pathway contributing to DNA synthesis and repair processes. This pathway connects with the one-carbon metabolism pathway where folate is an essential donor of carbons necessary for methylation reactions. In these pathways FOLR3 interacts with other folate receptors like FOLR1 and FOLR2 to maintain efficient folate transport and cellular homeostasis. Ensuring proper folate availability supports homocysteine metabolism which is another critical aspect of these pathways.

Associated diseases and disorders

FOLR3 relates notably to conditions such as cancer and inflammatory disorders. Cancer cells often exhibit elevated folate receptor expression including FOLR3 due to their increased requirement for folate in rapid cell proliferation.
