Transcription factor expressed by neural progenitor cells in specific regions of the embryonic neuroepithelium. Essential for the mammillary nuclei maintenance. Negatively regulates the proliferation of oligodendrocyte progenitors and promotes oligodendrocyte maturation. Also expressed in mammary glands, plays a role in lactation, controls development of mammary glands and the inferior colliculi of the midbrain in the central nervous system that regulates the milk-ejection reflex.

FOXB1 also known as Forkhead box protein B1 is a transcription factor. It typically weighs around 75 kDa. FOXB1 expression predominantly occurs in the brain where it regulates gene expression critical for development and function. This protein binds to DNA influencing the transcription of target genes which plays a role in development processes.
FOXB1 influences nervous system development and function acting as part of a complex that modulates transcriptional activity. Its role extends to maintaining cellular proliferation and differentiation. FOXB1's interaction with specific DNA elements shows its contribution to neurogenesis and axonal guidance which are essential for proper brain structure and function.
FOXB1 operates within the Hedgehog and Wnt signaling pathways which are essential for cell fate determination and growth in the brain. It works alongside proteins such as Gli in the Hedgehog signaling pathway to influence developmental processes. In the Wnt signaling pathway FOXB1 interacts with beta-catenin-related functions important for synaptic and neuronal activities in the brain.
FOXB1's involvement relates to neurodevelopmental disorders and glioblastoma. Altered FOXB1 expression or function can contribute to improper brain development leading to neurodevelopmental issues. In glioblastoma FOXB1 interacts with FoxG1 suggesting roles in tumor progression. Studying FOXB1 interactions offers insights into potential therapeutic targets for these conditions.
