FOXD1 also known as BF-2 acts as a transcription factor with a molecular weight of approximately 51 kDa. It binds to DNA to regulate gene expression influencing the transcription of down-stream genes. FOXD1 expression is most prominent in the kidney as well as in various regions of the brain and retina. The protein's action is important for development and differentiation processes in these tissues.
Biological function summary

The activity of FOXD1/BF-2 is central to organogenesis especially in the kidneys where it regulates nephron progenitor cells. It forms part of a gene regulatory complex that modulates the expression of other transcription factors necessary for organ development. Beyond organogenesis FOXD1 influences the function of the neural crest-derived tissues and contributes to patterning during embryonic development.

Pathways

The FOXD1/BF-2 protein integrates into the renin-angiotensin system impacting fluid homeostasis and blood pressure regulation. Additionally it participates in the Wnt signaling pathway an important pathway in cell fate determination. FOXD1 interacts with other proteins such as PRRX1 and GATA3 therefore solidifying its role in these signaling cascades.

FOXD1's dysfunction has links to renal dysplasia and hypertension. In renal dysplasia the improper expression of FOXD1 alters kidney development which affects renal function. Additionally FOXD1 alteration can relate to hypertensive disorders due to its role in the renin-angiotensin system. The interaction with PRRX1 highlights its involvement in these conditions causing ripple effects across the associated pathways.
