Activity summary

FOXL1 also known as Forkhead Box L1 is a transcription factor encoded [by the FOXL1 gene] and has a mass of approximately 38 kDa. This protein is part of the forkhead family of transcription factors which are characterized by a distinct forkhead domain. FOXL1 is expressed mainly in the mesenchymal cells of the gastrointestinal tract particularly in the stomach and the small intestine. Additionally it can be found in other tissues but at lower levels contributing to its role in the regulation of various cellular processes.

Biological function summary

The transcription factor FOXL1 plays a critical role in the development and differentiation of tissues within the digestive system. It functions as part of a complex that regulates mesenchymal-epithelial interactions during organogenesis particularly in the gut. FOXL1 influences the cell proliferation apoptosis and differentiation required for maintaining the homeostasis and architecture of the gastrointestinal epithelium. Its activity can modulate the expression of genes involved in the signaling pathways that are essential for tissue repair and development.

Pathways

FOXL1 impacts the hedgehog signaling and the Wnt/β-catenin pathway both of which are important to embryonic development and cell differentiation. In the hedgehog signaling pathway FOXL1 interacts with other proteins such as GLI transcription factors to modulate gene expression. Similarly the Wnt/β-catenin pathway involves FOXL1 in regulating processes linked to cell growth and differentiation in the intestinal environment often working with proteins like TCF7L2 to control target gene transcription.

Associated diseases and disorders

FOXL1 has connections to gastrointestinal cancers and congenital disorders affecting the digestive tract. Anomalies in FOXL1 expression or function can contribute to the development of stomach cancer where it may act together with the aberrant activation of FOXO3 and CTNNB1 proteins. Additionally disruptions in the regulatory roles of FOXL1 can lead to disorders such as duodenal atresia as it is fundamental in proper gastrointestinal development and maintenance. FOXL1's involvement in these diseases highlights its significance in understanding the molecular mechanisms underlying gastrointestinal pathologies.
