Activity summary

FOXQ1 also known as Forkhead box Q1 is a transcription factor that regulates gene expression. Its molecular weight is approximately 44 kDa. FOXQ1 expression occurs in various tissues including the lungs intestines and certain parts of the brain. This protein belongs to the Forkhead box (FOX) family which plays key roles in numerous cellular processes. These transcription factors share a similar DNA-binding domain enabling binding to specific sequences and control of target gene expression.

Biological function summary

FOXQ1 influences cell proliferation differentiation and survival. It does not form part of a larger complex though it interacts with other proteins for its function. In particular FOXQ1 can modulate epithelial-mesenchymal transition (EMT) an essential process in development and cancer metastasis. Through its role in EMT FOXQ1 impacts cellular plasticity and tumor progression thereby influencing how cells respond to growth signals.

Pathways

Connections involving FOXQ1 include the TGF-beta signaling and Wnt pathways. These pathways are critical in controlling cellular growth and differentiation functions. Within these pathways FOXQ1 interacts with proteins such as SMAD and beta-catenin which further transmit signals to the nucleus altering gene expression patterns. Such interactions position FOXQ1 as a pivotal component in facilitating cellular responses to environmental stimuli.

Associated diseases and disorders

FOXQ1 has implications in cancer and fibrotic diseases. In cancers such as colorectal and breast cancer abnormal FOXQ1 expression can lead to more aggressive tumor phenotypes and poor prognosis. FOXQ1 associates with proteins like E-cadherin and Vimentin during cancer progression influencing tumor invasiveness and metastasis. In fibrosis FOXQ1 dysregulation affects tissue repair by modifying fibroblast activity and matrix deposition showcasing its role in pathological tissue remodeling.

    Detection of human FOXQ1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from NCI-H520 cells prepared using NETN lysis buffer. Antibodies: Rabbit antiFOXQ1 recombinant monoclonal antibody [BLR230K] (A700-230) used for IP at 6 µl/mg lysate. FOXQ1 was also immunoprecipitated by a second antibody against a different epitope of FOXQ1 (BL-2269A-2C5). For blotting immunoprecipitated FOXQ1, A700-230 was used at 1:1000. Detection: Chemiluminescence with an exposure time of 30 seconds.
