Immunofluorescent analysis of HeLa cells labeling FRG1 with ab181083 at a 1/100 dilution. Primary incubation was followed by secondary antibody labeling with Alexa Fluor 555-conjugated goat anti-rabbit lgG (red - left panel). DAPI was used to stain the cell nuclear (blue - right panel).

FRG1 also known as FSHD Region Gene 1 is a protein with a molecular mass of approximately 30 kDa. It is expressed in a range of human tissues with notable expression in skeletal muscle and embryonic tissues. FRG1 plays a role in cellular activities related to the maintenance and function of muscle cells. Its involvement is linked to ribonucleoprotein complex assembly and it has potential implications in cellular localization of proteins.
Biological function summary

FRG1 is involved in processes related to muscle development and maintenance. It is a part of the spliceosome complex which plays an important role in mRNA splicing. Through its participation in this complex FRG1 helps regulate the maturation of pre-mRNA by removing introns. These activities are essential for accurate gene expression in cells particularly in tissues requiring precise regulation like muscle fibers.

Pathways

Four central components relate closely to FRG1's function. FRG1 is integrated into the mRNA processing and splicing pathways which are fundamental for synthesizing functional proteins. It interacts with proteins such as SMN1 which is associated with spinal muscular atrophy highlighting its connection to the ribonucleoprotein assembly pathway. Through these pathways FRG1 contributes to ensuring proper gene expression and cellular homeostasis.

Two prominent conditions involve FRG1: Facioscapulohumeral muscular dystrophy (FSHD) and certain myopathies. FSHD is associated with the abnormal expression of FRG1 leading to progressive muscle weakness. Other studies suggest FRG1 may also have interactions with DUX4 a protein implicated in FSHD pathogenesis. Understanding FRG1's role in these diseases provides insights into their molecular mechanisms supporting the development of therapeutic strategies.
