Frizzled 5 also known as FZD5 is a receptor that functions mechanically as a part of the Wnt signaling pathway. The Frizzled 5 protein has a molecular weight of around 75 kDa and is expressed in various tissues including the brain the eyes and the cardiovascular system. As a transmembrane receptor Frizzled 5 binds to Wnt ligands and activates intracellular signaling cascades mediating important cellular processes.
Biological function summary

Frizzled 5 participates in cellular communication and is part of the Wnt-frizzled protein complex. This complex plays a role in embryonic development cell proliferation and cell differentiation. Through its interaction with Wnt ligands Frizzled 5 can influence cell fate determination and contribute to the maintenance of stem cells. These activities are essential for development and tissue homeostasis.

Pathways

Knowledge of Frizzled 5 places it within both the canonical and non-canonical Wnt signaling pathways. In the canonical pathway Frizzled 5 associates with LRP5/6 co-receptors to stabilize β-catenin leading to gene transcription changes. In the non-canonical pathway it involves the activation of Ca2+ signaling. Proteins related to Frizzled 5 in these pathways include Dishevelled (DVL) and the Glycogen synthase kinase (GSK-3).

Research has linked Frizzled 5 to conditions like retinopathy of prematurity and colorectal cancer. In retinopathy Frizzled 5's interaction with Norrin—a protein related to eye vascular development—can lead to abnormal vascularization. In colorectal cancer alterations in the Wnt-Frizzled signaling are implicated with β-catenin accumulating as a consequence. The modification in these signaling pathways hints at the potential for therapeutic interventions targeting Frizzled 5.
