Frizzled 8 also known as FZD8 forms part of the Frizzled family of proteins which serve as receptors for the Wnt signaling pathway. This protein with a mass of approximately 70 kDa contains a characteristic cysteine-rich domain. FZD8 mainly localizes to the cell membrane and prominently expresses in tissues such as the brain heart and placenta. Its structure includes seven transmembrane domains which contributes significantly to its role in signal transduction.
Biological function summary

FZD8 influences cellular communication within the Wnt signaling pathway impacting processes like cell proliferation and fate determination. FZD8 often associates with other proteins in complex formations particularly when assembling with Wnt ligands on the cell surface. It plays an important role in embryonic development and tissue homeostasis where precise cellular signaling is necessary for maintaining the balance between cell growth and programmed cell death.

Pathways

FZD8 interacts most prominently within the canonical Wnt/β-catenin signaling pathway. It works closely with the Dishevelled (DVL) protein facilitating signal transfer from the cell surface to the cytoplasm and beyond. This involvement tightly links FZD8 to many developmental processes and cellular communication networks. Additionally FZD8 is also implicated in the non-canonical Wnt pathways demonstrating its versatility in various cellular signaling contexts.

FZD8 is associated with conditions such as cancer and neurodegenerative diseases. Dysregulation of FZD8 may contribute to abnormal cell proliferation as observed in certain cancers when improper Wnt signaling causes uncontrolled cellular growth. Additionally its connection with Alzheimer's disease highlights the importance of FZD8 in neuronal maintenance and function. Abberations in FZD8 or its signaling partners such as β-catenin may lead to pathogenesis in these diseases.

Target data

Receptor for Wnt proteins. Component of the Wnt-Fzd-LRP5-LRP6 complex that triggers beta-catenin signaling through inducing aggregation of receptor-ligand complexes into ribosome-sized signalosomes. The beta-catenin canonical signaling pathway leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes. A second signaling pathway involving PKC and calcium fluxes has been seen for some family members, but it is not yet clear if it represents a distinct pathway or if it can be integrated in the canonical pathway, as PKC seems to be required for Wnt-mediated inactivation of GSK-3 kinase. Both pathways seem to involve interactions with G-proteins. May be involved in transduction and intercellular transmission of polarity information during tissue morphogenesis and/or in differentiated tissues. Coreceptor along with RYK of Wnt proteins, such as WNT1.
