Supplementary information

Frizzled 9 also known as FZD9 or the frizzled class receptor 9 acts as a receptor for Wnt proteins. This protein is a member of the Frizzled family involved in the Wnt signaling pathway. Frizzled 9 has an approximate mass of 67 kDa. Researchers have found that it expresses in various tissues particularly in the brain lungs and bone. Its expression pattern indicates a role in both development and disease processes.
Biological function summary

Frizzled class receptor 9 functions in cellular communication and development. It does not operate alone but often forms complexes with co-receptors like LDL receptor-related protein 5/6 (LRP5/6). Through these complexes it transmits signals important for various cellular functions such as proliferation differentiation and migration. Frizzled 9’s signaling affects cellular processes contributing significantly to maintaining normal cellular homeostasis.

Pathways

Frizzled receptor 9 participates in the Wnt/β-catenin signaling and the planar cell polarity (PCP) pathway. In the Wnt/β-catenin pathway frizzled 9 interacts with proteins such as β-catenin and Dishevelled (Dvl) leading to gene expression changes. In the PCP pathway frizzled 9 contributes to the regulation of cellular orientation within tissues. Through these pathways frizzled 9 plays a role in cell fate determination and tissue patterning making it an important component in organismal development and disease.

Alterations in frizzled 9 function link to osteoporosis and cancer. Its expression levels and signaling efficiency can impact bone density connecting it to LRP5/6 in osteoporosis. Abnormal frizzled 9 signaling may also influence cancer development as disruptions in the Wnt pathway are known to affect cell proliferation and tumor growth. Understanding frizzled 9’s interplay with these proteins provides insight into its role in disease mechanisms and potential therapeutic implications.

Target data

Receptor for WNT2 that is coupled to the beta-catenin canonical signaling pathway, which leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes (By similarity). Plays a role in neuromuscular junction (NMJ) assembly by negatively regulating the clustering of acetylcholine receptors (AChR) through the beta-catenin canonical signaling pathway (By similarity). May play a role in neural progenitor cells (NPCs) viability through the beta-catenin canonical signaling pathway by negatively regulating cell cycle arrest leading to inhibition of neuron apoptotic process (PubMed : 27509850). During hippocampal development, regulates neuroblast proliferation and apoptotic cell death. Controls bone formation through non canonical Wnt signaling mediated via ISG15. Positively regulates bone regeneration through non canonical Wnt signaling (By similarity).
