FRK also known as FYN-related kinase or RAK is a non-receptor tyrosine kinase with a molecular mass of approximately 56 kDa. This protein is predominantly expressed in epithelial tissues including those of the breast kidney and liver. FRK displays substrate specificity and catalytic function by phosphorylating specific tyrosine residues on target proteins which influences cellular processes such as proliferation and differentiation.
Biological function summary

This kinase contributes to the regulation of cell cycle progression and apoptosis. FRK acts solo as well as part of larger signal transduction complexes. It plays a vital role in limiting uncontrolled cellular growth which is particularly significant in maintaining tissue homeostasis. FRK regulates various molecular interactions essential for cell adhesion and communication adding another layer of control over cell physiology.

Pathways

FRK integrates into signaling pathways like the PI3K/AKT and MAPK pathways. These pathways are critical for mediating responses to growth signals and stress conditions. FRK's activity modulates protein interactions with key molecules such as p53 and PTEN influencing downstream effects that control cell cycle checkpoints and apoptosis. By interacting with these pathways FRK helps to balance cell proliferation and survival in response to extracellular cues.

FRK has implications in cancer development particularly in breast cancer. Altered FRK expression or activity has been associated with tumor progression and resistance to treatment. FRK interacts with proteins such as BRCA1 contributing to its role in DNA damage response and tumor suppression. Additionally FRK's aberrant activity links to certain forms of renal disorders where it impacts cellular responses in renal tissue often through modulation of E-cadherin and β-catenin in epithelial cells.

Non-receptor tyrosine-protein kinase that negatively regulates cell proliferation. Positively regulates PTEN protein stability through phosphorylation of PTEN on 'Tyr-336', which in turn prevents its ubiquitination and degradation, possibly by reducing its binding to NEDD4. May function as a tumor suppressor.
