Product details

ab317352 is a BSA and Azide free antibody supplied in an unconjugated format and it is suitable for sandwich ELISAs to quantify FRS2. The recommended pair for sandwich ELISA is:

Capture: ab317351, FRS2 Capture Antibody (unconjugated)

Detector: ab317352, FRS2 + FSR3 Detector Antibody (unconjugated)

The reference range value is 19.53-2500 pg/mL.

The recommended antibody orientation is based on internal optimization for ELISA based assays. Antibody orientation is assay dependent and needs to be optimized for each assay type. Please note that the range provided for this antibody is only an estimation based on the performance of the product using the recommended antibody pair. Performance of the antibody pair will depend on the specific characteristics of your assay. We guarantee the product works in sandwich ELISA, but we do not guarantee the sensitivity or dynamic range of the antibody in your assay.

Our carrier-free formats are supplied in a buffer free of BSA, sodium azide and glycerol for higher conjugation efficiency. The antibodies are provided at an approximate concentration of 1 mg/ml as measured by the protein A280 method. Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with <1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Supplementary information

The FRS2 and FRS3 proteins also known as Fibroblast Growth Factor Receptor Substrate 2 and 3 serve as adaptor proteins in signal transduction processes. FRS2 with a mass of approximately 60 kDa and FRS3 play key roles in linking cell surface receptors to downstream signaling pathways. They are ubiquitously expressed but have higher concentrations in certain tissues like the brain and heart. These proteins have modular structures that contain phosphotyrosine-binding domains and SH2 domains which are involved in protein-protein interactions.
Biological function summary

The adaptor proteins FRS2 and FRS3 mediate signals from receptor tyrosine kinases notably the Fibroblast Growth Factor Receptors (FGFRs). Through their action they facilitate the assembly of signaling complexes necessary for cellular proliferation differentiation and survival. FRS2 and FRS3 activate important downstream effectors like the Ras/MAPK and PI3K/AKT pathways. They not only recognize activated receptors but also recruit other adaptors and signaling molecules to form complex networks.

Pathways

The proteins FRS2 and FRS3 are integral to the FGFR and ERK/MAPK signaling pathways. In these pathways they are involved in transmitting external growth signals to the cell's interior leading to varied biological responses. They interact with other pathway components including GRB2 and SOS which further propagate the signal cascade. This places FRS2 and FRS3 at a central point in controlling cellular responses to external stimuli.

Mutations or malfunctions in FRS2 and FRS3 have been implicated in several conditions notably cancer and developmental disorders. Aberrant signaling through these adaptors can lead to uncontrolled cell proliferation contributing to oncogenesis. Additionally they have connections to developmental defects where improper signaling affects organogenesis and tissue formation. These proteins interact with key components such as FGFRs which are known to be altered in these disease states further linking FRS2 and FRS3 to the pathological processes.

Target data

Adapter protein that links activated FGR and NGF receptors to downstream signaling pathways. Plays an important role in the activation of MAP kinases and in the phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase, in response to ligand-mediated activation of FGFR1. Modulates signaling via SHC1 by competing for a common binding site on NTRK1.
