Supplementary information

FTSJ3 also known as mitochondrial ribosomal RNA methyltransferase 3 is an enzyme with a mass of approximately 84 kDa. The protein is part of the FtsJ/hEN1 family and plays a role in RNA modification by adding methyl groups to ribosomal RNA (rRNA) impacting ribosome assembly and function. FTSJ3 is ubiquitously expressed with high levels in tissues like the heart liver and skeletal muscle indicating its importance for metabolic activity and protein synthesis.
Biological function summary

FTSJ3 functions as an rRNA methyltransferase that is important for the maintenance of mitochondrial ribosome stability. It is a component of the mitoribosome complex which is responsible for mitochondrial protein synthesis. This activity is essential for cellular energy production as it directly affects the translation of mitochondrial-encoded genes required for oxidative phosphorylation.

Pathways

FTSJ3 plays a role in mitochondrial gene expression pathways and its activity affects the oxidative phosphorylation pathway a fundamental process for ATP production. This protein works alongside related proteins like METTL12 which has complementary roles in RNA modification ensuring the stability and efficiency of mitochondrial translation machinery.

Disruptions in FTSJ3 function associate with mitochondrial diseases that involve energy deficiencies such as Leigh syndrome and mitochondrial encephalomyopathy. The protein's dysfunction can also impact neurological disorders due to its critical role in energy production connecting it to proteins like GFM1 and MRPS7 which are also involved in mitochondrial translation and whose dysfunctions similarly contribute to mitochondrial pathologies.

Target data

RNA 2'-O-methyltransferase involved in the processing of the 34S pre-rRNA to 18S rRNA and in 40S ribosomal subunit formation.. (Microbial infection) In case of infection by HIV-1 virus, recruited to HIV-1 RNA and catalyzes 2'-O-methylation of the viral genome, allowing HIV-1 virus to escape the innate immune system (PubMed : 30626973). RNA 2'-O-methylation provides a molecular signature for discrimination of self from non-self and is used by HIV-1 to evade innate immune recognition by IFIH1/MDA5 (PubMed : 30626973). Mediates methylation of internal residues of HIV-1 RNA, with a strong preference for adenosine (PubMed : 30626973). Recruited to HIV-1 RNA via interaction with TARBP2/TRBP (PubMed : 30626973).
