Supplementary information

FUT10 also known as fucosyltransferase 10 is an enzyme responsible for transferring fucose to glycoproteins and glycolipids. This enzyme has a predicted molecular mass of approximately 50 kDa. Expression of FUT10 occurs in various tissues including the brain and liver indicating its broad functional role across different cell types. Its activity impacts the synthesis and modification of cell surface molecules which can influence cell-cell communication and signal transduction.
Biological function summary

FUT10 plays a significant role in the process of glycosylation where it is involved in the addition of fucose moieties to substrates. It functions independently and does not form part of any known larger protein complex. FUT10 modifies glycoproteins that can alter cellular interactions and immune recognition affecting processes such as cell adhesion and migration. Glycoproteins fucosylated by FUT10 include cell adhesion molecules some of which may serve in important developmental and immune response roles.

Pathways

FUT10 interacts with systems of glycan biosynthesis and metabolism. It participates in the N-glycan biosynthesis pathway working together with other enzymes such as FUT8 which also adds fucose residues to complexes. FUT10's activity in this pathway helps control the structure of glycans which can modulate signaling pathways involved in cellular growth and differentiation.

Aberrations in FUT10 function have links to cancer and immune system disorders. Changes in the glycosylation patterns influenced by FUT10 can arise in cancer cells affecting cell recognition and metastasis. The enzyme also connects to immune system dysfunction as altered fucosylation patterns can impact immune cell signaling pathways. FUT10's role in these conditions additionally intersects with proteins like E-selectin which binds select fucosylated structures involved in immune responses and cancer progression.

Target data

Predominantly fucosylates the innermost N-acetyl glucosamine (GlcNAc) residue in biantennary N-glycan acceptors. Postulated to generate core alpha(1->3)-fucose epitope within the chitobiose unit of biantennary N-glycans, providing for a recognition signal to reorient aberrantly folded glycoproteins for degradation (PubMed : 19088067). Involved in biosynthesis of Lewis X-carrying biantennary N-glycans that regulate neuron stem cell self-renewal during brain development (By similarity).. Isoform 1. Catalyzes the transfer of fucosyl moiety from GDP-beta-L-fucose to the innermost GlcNAc residue in biantennary N-glycan acceptors. Does not fucosylate GlcNAc within type 2 lactosamine unit.. Isoform 4. Catalyzes the transfer of fucosyl moiety from GDP-beta-L-fucose to the innermost GlcNAc residue in biantennary N-glycan acceptors. Does not fucosylate GlcNAc within type 2 lactosamine unit.. Isoform 5. Catalyzes the transfer of fucosyl moiety from GDP-beta-L-fucose to the innermost GlcNAc residue in biantennary N-glycan acceptors. Does not fucosylate GlcNAc within type 2 lactosamine unit.
