FUT6 also known as fucosyltransferase 6 is an enzyme with a molecular mass of approximately 45 kDa. This enzyme mechanically transfers fucose a type of sugar to N-glycans found on cell surfaces. FUT6 achieves this through an α13 or α14-glycosidic linkage. Expression of FUT6 occurs mainly in the liver and gastrointestinal tract indicating its role in tissues involved in metabolism and digestion.
Biological function summary

This enzyme plays a role in the biosynthesis of the Lewis blood group antigens which are important for cell-cell interactions. These interactions occur during processes like immune responses and cellular differentiation. FUT6 functions in this context without forming part of larger protein complexes. Its activity directly affects the modification of oligosaccharides to present selectin ligands which facilitates cellular adhesion.

Pathways

FUT6 is an essential player in the fucosylation pathway impacting glycoprotein and glycolipid metabolism. It interacts closely with other members of the fucosyltransferase family such as FUT4 to modulate synthesis of sialyl Lewis x epitopes. These pathways are instrumental in regulating leukocyte extravasation during immune surveillance and inflammation.

FUT6 shows significant relevance to inflammatory diseases and certain types of cancer. Abnormal expression of this enzyme has links to colorectal cancer progression due to altered glycan structures on the cell surface. Additionally FUT6 in connection with other fucosyltransferases like FUT3 relates to inflammatory bowel disease where changes in glycosylation patterns might contribute to pathogenesis.

Catalyzes the transfer of L-fucose, from a guanosine diphosphate-beta-L-fucose, to the N-acetyl glucosamine (GlcNAc) of a distal alpha2,3 sialylated lactosamine unit of a glycoprotein- or a glycolipid-linked sialopolylactosamines chain or of a distal or internal lactosamine unit of a neutral glycoprotein- or a glycolipid-linked polylactosamines chain through an alpha-1,3 glycosidic linkage and participates in surface expression of the sialyl Lewis X (sLe(x)), Lewis X (Le(x)) and non sialylated VIM2 determinants (PubMed : 10728707, PubMed : 1339443, PubMed : 1520296, PubMed : 17604274, PubMed : 29593094, PubMed : 7650030, PubMed : 9363434, PubMed : 9451035). Also fucosylates a polylactosamine substrate having a 6 sulfate modification at the GlcNAc moiety and gives rise to sialyl and non-sialyl 6-sulfo lewis X (PubMed : 10728707). Does not have activity towards type 1 ((Galbeta1-3GlcNAc)) and H-type 1 chain (Fucalpha1-2Galbeta1-3GlcNAc) acceptors substrates (PubMed : 1339443, PubMed : 17604274, PubMed : 9363434).. Isoform 2. Does not have alpha(1,3)-fucosyltransferase activity.
