Supplementary information

The protein FXR2 also known as Fragile X Mental Retardation Syndrome-Related Protein 2 is part of the fragile X-related family of proteins. It has a molecular mass of approximately 78 kDa. This protein is expressed in various tissues with higher levels in the brain and testes. FXR2 acts primarily as an RNA-binding protein and has functional RNA transport and translational repression roles. The protein contains several motifs including a KH domain and RGG boxes which aid its interaction with RNA.
Biological function summary

FXR2 is a part of a larger complex that includes homologous proteins like FMRP and FXR1. These proteins cooperate in the regulation of mRNA dynamics in neurons thereby playing essential roles in synaptic function and plasticity. They affect the translation of specific mRNAs and modulate the structure of dendritic spines. The FXR2 protein is important in the proper development of neurological architecture and cognitive processes.

Pathways

FXR2 participates in the mTOR signaling pathway and the regulation of dendritic translation. The mTOR pathway is vital for cell growth and protein synthesis critical in neural activity and development. FXR2 works alongside related proteins such as FMR1 and FXR1 within these pathways and these interactions modulate ribosome biogenesis and activity. FXR2 interactions importantly facilitate enhanced responses to synaptic activities.

FXR2 influences conditions like Fragile X syndrome and autism spectrum disorders. Aberrations in FXR2 expression or function disrupt neuronal development and synaptic plasticity and are linked to cognitive impairments. FXR2 shares close connections with proteins FMR1 and CYFIP1 within these disorders suggesting a broader network of protein interactions critical for normal neurological function.

Target data

mRNA-binding protein that acts as a regulator of mRNAs translation and/or stability, and which is required for adult hippocampal neurogenesis (By similarity). Specifically binds to AU-rich elements (AREs) in the 3'-UTR of target mRNAs (By similarity). Promotes formation of some phase-separated membraneless compartment by undergoing liquid-liquid phase separation upon binding to AREs-containing mRNAs : mRNAs storage into membraneless compartments regulates their translation and/or stability (By similarity). Acts as a regulator of adult hippocampal neurogenesis by regulating translation and/or stability of NOG mRNA, thereby preventing NOG protein expression in the dentate gyrus (By similarity).
