FXYD6 also known as phospholemman-like protein (Plp) is a member of the FXYD family that regulates ion transport. This protein has a mass of around 9 kDa and demonstrates specificity in modulating sodium-potassium (Na+/K+) ATPase activity. The expression of FXYD6 is quite widespread but appears higher in tissues such as brain and kidney suggesting its important roles in these organs' physiological functions. It influences ion balance and potentially affects cellular excitability and fluid transport through its modulation of ion channels and pumps.
Biological function summary

FXYD6 impacts multiple cellular mechanisms mainly through its association with Na+/K+ ATPase complex. It influences ion gradients across the plasma membrane an essential factor in maintaining cell volume and resting membrane potential. Interactions within this complex suggest FXYD6 plays an important part in fine-tuning ion pump efficiency affecting the overall cellular ionic homeostasis. Its precise role within the ATPase complex is important for adapting the function of these pumps to specific physiological conditions.

Pathways

FXYD6 integrates into ion transport and signal transduction pathways particularly those involving Na+/K+ ATPase. It cooperates with other FXYD proteins like FXYD2 in adjusting ion pump functionality ensuring cells respond adequately to physiological stimuli. Associates with and regulates the activity of the sodium/potassium-transporting ATPase (NKA) which catalyzes the hydrolysis of ATP coupled with the exchange of Na(+) and K(+) ions across the plasma membrane. Reduces the apparent affinity for intracellular Na(+) with no change in the apparent affinity for extracellular K(+) (PubMed : 33231612). In addition to modulating NKA kinetics, may also function as a regulator of NKA localization to the plasma membrane (By similarity).
