Activity summary

GAD1b often simply referred to as GAD1 or glutamate decarboxylase 1 is an enzyme that catalyzes the conversion of glutamate to gamma-aminobutyric acid (GABA). This enzyme is a part of the GAD family and has a molecular mass of approximately 67 kDa. GAD1b is primarily expressed in the brain particularly within neurons in the cortex and hippocampus where it plays an essential role in neurotransmitter regulation. This expression is important for maintaining the balance between excitatory and inhibitory signals in the nervous system.

Biological function summary

Glutamate decarboxylase 1 functions as an important regulator of the inhibitory neurotransmitter GABA. GAD1b exists as part of a functional enzyme complex which is involved in neurotransmitter synthesis. This enzymatic activity is fundamental to inhibitory signal transmission and synaptic plasticity which are essential processes for cognitive functions such as learning and memory.

Pathways

Glutamate decarboxylase 1 is an important component in the GABAergic signaling pathway. This pathway involves neurotransmitter production synaptic vesicle release and receptor binding which significantly affects neuronal excitability and mood regulation. GAD1b is linked with other proteins such as GAD2 and various GABA receptor subunits which collectively modulate synaptic inhibition across different brain regions.

Associated diseases and disorders

Disruptions in GAD1b expression or function can lead to neurological conditions like epilepsy and schizophrenia. These disorders often involve an imbalance in excitatory and inhibitory neurotransmission highlighting the importance of GAD1b's role in producing GABA. Other proteins including GAD2 also play a role in these diseases often contributing to the underlying pathology through their participation in GABA synthesis and regulation pathways.
