This data was developed using ab305101, the same antibody clone in a different buffer formulation. Immunohistochemical analysis of paraffin-embedded Human testis tissue labeling GAK with ab305101 at 1/50 (9.9 ug/ml) followed by a ready to use Goat Anti-Rabbit IgG H&L (HRP polymer). Positive staining on human testis (PMID : 9299234). The section was incubated with ab305101 at 4°C overnight. Counterstained with Hematoxylin. Secondary antibody only control : Secondary antibody is a ready to use Goat Anti-Rabbit IgG H&L (HRP polymer). Heat mediated antigen retrieval was performed using ab93684 (Tris/EDTA buffer, pH 9.0)

This data was developed using ab305101, the same antibody clone in a different buffer formulation. Blocking and diluting buffer and concentration : 5% NFDM/TBST In Western blot, ab305101 was shown to bind specifically to GAK. A band was observed at 160 kDa in wild-type 293T cell lysates with no signal observed at this size in GAK knockout cell line ab266417 (knockout cell lysate ab263210). Exposure time : 48 seconds

Target data

Associates with cyclin G and CDK5. Seems to act as an auxilin homolog that is involved in the uncoating of clathrin-coated vesicles by Hsc70 in non-neuronal cells. Expression oscillates slightly during the cell cycle, peaking at G1 (PubMed : 10625686). May play a role in clathrin-mediated endocytosis and intracellular trafficking, and in the dynamics of clathrin assembly/disassembly (PubMed : 18489706).
