GALNT8 also known as polypeptide N-acetylgalactosaminyltransferase 8 is an enzyme that participates in the initial glycosylation steps transferring N-acetylgalactosamine to serine and threonine residues on proteins. This enzyme has a molecular mass of approximately 70 kDa. It is expressed in various tissues including the brain and liver. The enzyme belongs to the family of GALNTs which are key players in O-glycosylation a common form of post-translational modification.
Biological function summary

GALNT8 contributes to the synthesis of mucin-type O-glycans which are essential for the formation of mucous barriers and modulation of protein functions. The enzyme does not usually form part of large protein complexes but it can act in a sequential manner with other glycosyltransferases. This function is needed for proper protein stability and activity and for protecting proteins from proteolysis.

Pathways

GALNT8 participates in the O-glycan biosynthesis pathway. This pathway involves enzymes like GALNT2 and GALNT3 that help add sugar moieties to protein substrates. Additionally GALNT8 influences cell signaling and cellular interactions because glycosylation changes protein interactions and signaling receptor functions.

GALNT8 has links to conditions such as cancer and neurological disorders. Alterations in glycosylation patterns driven by GALNT8 affect tumor cell behavior including adhesion invasion and metastasis. In cancer GALNT8 may interact with proteins like MUC1 a mucin protein often overexpressed in tumors. Aberrant expression of GALNT8 may also influence neurological diseases where glycosylation changes impact neuronal communication and function.
