Multiplex immunohistochemistry analysis of formalin/PFA-fixed paraffin-embedded rat dorsal root ganglia staining GALR2 with ab271019 at a 1/2000 dilution and ab177487 anti-NeuN used at 1/300 dilution.

Panel A : merged staining of anti-GALR2 (green; Opal™520) and anti-NeuN (magenta; Opal™570) on Rat dorsal root ganglia.

Panel B : anti-GALR2 stained on cytoplasm of neurons.

Panel C : anti-NeuN stained on nucleus and cytoplasm of neurons.

Panel D : Nuclear DNA was labeled with DAPI (shown in blue).

The section was incubated in two rounds of staining : in the order of ab271019 and ab177487 for 30 mins at room temperature. Each round was followed by a separate fluorescent tyramide signal amplification system.

The immunostaining was performed on a Leica Biosystems BOND® RX instrument with an Opal™ 4-color kit. Image acquisition was performed with Leica SP8 confocal microscope.

Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins.

Immunohistochemical analysis of 4% PFA-fixed, 0.2% Triton X-100 permeabilized frozen Rat cerebral cortex tissue labeling GALR2 with ab271019 at 1/100 (5.17 ug/ml) dilution followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 dilution (Green). Cytoplasmic staining on rat cerebral cortex is observed. The nuclear counterstain was DAPI (Blue).

Secondary antibody control : Secondary antibody is ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 dilution.

Heat mediated antigen retrieval using sodium citrate buffer (10mM citrate pH 6.0 + 0.05% Tween-20).

GALR2 also known as Galanin receptor type 2 is a G protein-coupled receptor with a molecular mass of around 38 kDa. This receptor is expressed in several tissues including the central nervous system peripheral nervous system and various peripheral organs. GALR2 interacts with the neuropeptide galanin which modulates several physiological functions. The receptor signals through different classes of G proteins leading to the activation of intracellular second messengers.
Biological function summary

GALR2 influences numerous processes essential for maintaining homeostasis. It mediates cellular responses to stress appetite and mood through interaction with its ligand galanin. Though GALR2 does not form part of a larger receptor complex it plays a central role in its signaling pathways. Researchers recognize its function in promoting neurogenesis and modulating neurotransmitter release impacting several neural circuits.

Pathways

GALR2 plays roles in the MAPK/ERK pathway and the PI3K/AKT pathway. These pathways are essential for cell survival differentiation and proliferation. GALR2 interacts closely with other receptors such as GALR1 and GALR3 sharing similar downstream effects while maintaining distinct cellular responses. GALR2 has associations with depression and epilepsy.
