Supplementary information

Ganglioside GD2 also called G2a GD 2 or G2a uno is a glycolipid present on the outer layer of the cell membrane. It is a member of the ganglioside family a class of sialic acid-containing glycosphingolipids important in cellular interaction. GD2 has a molecular mass of approximately 1557 Daltons. Expression of GD2 is observed significantly in peripheral tissues like the central nervous system and on the surface of some cancer cells. Its high expression on select tumor types makes it a significant focus for monoclonal antibody development including anti-GD2 antibodies.
Biological function summary

Gangliosides like GD2 participate in cell-cell communication and modulation of cell signaling. GD2 is not part of a larger protein complex; rather it is integrated into the lipid bilayer mediating signaling processes and influencing cell adhesion migration and proliferation. The structure of GD2 allows it to engage in interactions impacting cellular behavior and phenotype especially in neuronal and immune system environments.

Pathways

Ganglioside GD2 plays a role in membrane microdomains known as lipid rafts which participate in signal transduction pathways and cellular trafficking. It contributes to the Wnt signaling pathway affecting cellular proliferation and differentiation. GD2 associates with other proteins such as Dishevelled in the Wnt pathway modulating downstream effects. Through these interactions it influences cellular responses necessary for development and tissue homeostasis.

Ganglioside GD2 is implicated in cancers particularly neuroblastoma and melanoma. Its association with cancer progression makes it a target for immunotherapy with therapies using anti-GD2 antibodies like dinutuximab approved for clinical use. This therapeutic approach leverages the presence of GD2 to target tumor cells. GD2's high expression on tumor cells and low expression on normal tissues makes it attractive for targeted therapies offering a route to distinguish between cancer cells and healthy cells in treatment strategies.
