GATA2 was immunoprecipitated from 0.35 mg K-562 (Human chronic myelogenous leukemia lymphoblast) cell lysate 10 μg with ab109241 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab109241 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab109241).

Chromatin was prepared from K-562 (Human chronic myelogenous leukemia lymphoblast) cells according to the Abcam Dual-X-ChIP protocol. Cells were fixed with 1.5 mM EGS for 30mins and then 1% formaldehyde for 10min.

The ChIP was performed with 25 µg of chromatin, 5 µg of ab109241 (red), or 5 µg of rabbit normal IgG ab172730 (gray) and 20 µl of Protein A/G sepharose beads. The immunoprecipitated DNA was quantified by real time PCR ( Sybr green approach).

GATA2 also known as GATA binding protein 2 is a transcription factor with a molecular mass of approximately 50 kDa. It plays an important mechanical role in regulating gene expression by binding to the DNA sequence "GATA". GATA2 is expressed in various tissues including hematopoietic cells the nervous system and the endothelium. It influences cell fate decisions impacting the development and function of multiple tissues and cell types.
Biological function summary

GATA2 governs hematopoiesis and vascular development. It operates within multiprotein complexes to control the transcription of genes necessary for the development and differentiation of hematopoietic stem cells. GATA2's presence aids in maintaining normal functions of these cells ensuring they proliferate and differentiate correctly. Its regulatory role extends to the vascular system where GATA2 controls endothelial cell function and blood vessel formation.
