Purification notes

ab138451 was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific phosphopeptide. The antibody against non-phosphopeptide was removed by chromatography using non-phosphopeptide corresponding to the phosphorylation.

Activity summary

GATA2 also known as GATA binding protein 2 is a transcription factor with a molecular mass of approximately 50 kDa. It plays an important mechanical role in regulating gene expression by binding to the DNA sequence "GATA". GATA2 is expressed in various tissues including hematopoietic cells the nervous system and the endothelium. It influences cell fate decisions impacting the development and function of multiple tissues and cell types.

Biological function summary

GATA2 governs hematopoiesis and vascular development. It operates within multiprotein complexes to control the transcription of genes necessary for the development and differentiation of hematopoietic stem cells. GATA2's presence aids in maintaining normal functions of these cells ensuring they proliferate and differentiate correctly. Its regulatory role extends to the vascular system where GATA2 controls endothelial cell function and blood vessel formation.

Pathways

Hematopoiesis and vascular development significantly involve GATA2. It participates in the Notch signaling pathway influencing cell differentiation and proliferation along with other transcription factors like GATA1 and FOG1. The pathway interplay facilitates normal blood cell production and proper vascularization emphasizing its important role in cellular development processes.

Associated diseases and disorders

Defective GATA2 function links to immunodeficiency syndromes and acute myeloid leukemia (AML). Mutations in the GATA2 gene disrupt normal blood cell development often resulting in increased susceptibility to infections and hematologic malignancies. It interacts with proteins such as GATA1 in these pathological states highlighting its significance in both immune function and malignant transformation.
