Protein involved in mitophagy by facilitating recruitment of the autophagy machinery required for clearance of damaged mitochondria (PubMed:30982665). Accumulates on the mitochondria surface in response to mitochondrial depolarization and acts as a 'eat me' signal by recruiting proteins involved in selective autophagy, such as autophagy receptors (CALCOCO2/NDP52, NBR1, SQSTM1/p62, TAX1BP1 and WDFY3/ALFY) and ATG8 family proteins (MAP1LC3A, MAP1LC3B, MAP1LC3C, GABARAP, GABARAPL1 and GABARAPL2) (PubMed:30982665).

Activity summary

The GBAS protein also known as Golgi apparatus-specific brefeldin A-resistance guanine nucleotide exchange factor 1 (GBF1) is a significant protein in cellular mechanics. GBAS has an approximate molecular weight of 200 kDa. You can find its expression mostly in tissues with high cellular turnover rates such as epithelial tissues and certain types of immune cells. This protein mainly functions within the Golgi apparatus where it plays an important role in regulating vesicular trafficking.

Biological function summary

GBAS contributes to essential cellular processes by participating in the activation of ADP-ribosylation factors (ARFs) as part of a larger protein complex. The protein helps control cargo sorting and lipid composition within the Golgi influencing the secretory pathway. Because of its role in regulating ARFs GBAS is vital in the maintenance of membrane trafficking and cytoskeletal organization impacting cell morphology and function.

Pathways

GBAS is deeply involved in the secretory and endocytic pathways impacting cellular homeostasis. It interacts with proteins like ARF4 and ARF5 ensuring efficient vesicle formation and transport. GBAS acts as a bridge within pathways that manage protein and lipid modifications prior to cell surface delivery or lysosomal degradation.

Associated diseases and disorders

Research has linked GBAS activity to neurodegenerative disorders and certain cancers given its role in vesicular trafficking. Its dysregulation in neurodegenerative diseases often points to malfunction in ARF-dependent pathways that affect neuron maintenance. In cancers alterations in GBAS function can lead to changes in cellular growth and differentiation frequently involving proteins like ARF1 highlighting its potential as a biomarker or therapeutic target.
