Ghrelin also known as Ghrelin-28 is a peptide hormone with a mass of approximately 3.3 kDa. It functions as an important regulator of appetite and energy balance. Ghrelin is secreted mainly by the stomach's enteroendocrine cells particularly the P/D1 cells in humans. In addition it is present in smaller amounts in the pancreas intestines and hypothalamus. The peptide is known for its ability to stimulate the release of growth hormone from the anterior pituitary gland acting via the growth hormone secretagogue receptor (GHS-R).
Ghrelin plays a significant role in hunger signaling and energy homeostasis. It is not typically part of a protein complex but acts as a ligand for GHS-R. By binding to the receptor in the brain especially in the hypothalamus Ghrelin promotes food intake and can influence body weight regulation. The hormone also affects glucose metabolism and has impacts on cardiovascular functions. Its presence in the gastrointestinal tract suggests involvement in gastric motility and acid secretion.
Ghrelin is notably involved in the GH/IGF-1 axis and the central nervous system pathways that regulate appetite.
