Supplementary information

GK2 also known as Glycerol Kinase 2 is primarily an enzyme that catalyzes the phosphorylation of glycerol to form glycerol-3-phosphate. This enzyme activity is essential in lipid metabolism. The protein is relatively small with a molecular mass of about 60 kDa. Expression of GK2 occurs mostly in the testes although it is also found in other tissues indicating a specific but not exclusive role in male physiology.
Biological function summary

GK2 contributes significantly to the metabolic processes involving glycerol and energy storage. It functions as part of an enzymatic sequence for transforming glycerol into a form usable for energy production. While GK2 is not traditionally associated with complex structures it interacts closely with other enzymes in the glycerol-3-phosphate pathway to maintain its function. This interaction ensures effective regulation of glycerol levels in cells aiding metabolic balance.

Pathways

GK2 plays a significant role in lipid metabolism and energy homeostasis. The glycerol-3-phosphate shuttle utilizes GK2 to facilitate the transfer of reducing equivalents into mitochondria linking it to oxidative phosphorylation. Proteins such as glycerol-3-phosphate dehydrogenase are related to GK2 within these pathways providing a tight regulation and ensuring the efficient flow of metabolites through the cycle.

GK2 dysfunction appears connected to metabolic syndromes and fertility issues. Variations in GK2 expression and activity have been observed in cases of metabolic syndrome impacting overall lipid levels. In disorders related to male fertility GK2 interactions with proteins like glycerol-3-phosphate dehydrogenase could influence energy supply affecting spermatogenesis and fertility outcomes.

Target data

Key enzyme in the regulation of glycerol uptake and metabolism. Essential for male fertility and sperm mitochondrial sheath formation (By similarity). Required for proper arrangement of crescent-like mitochondria to form the mitochondrial sheath during spermatogenesis (By similarity). Can induce mitochondrial clustering through interactions with PLD6 and up-regulation of phosphatidic acid synthesis in the mitochondria (PubMed : 28852571).
