IHC image of Glucose Transporter GLUT4 staining in a section of formalin-fixed paraffin-embedded normal human heart* performed on a Leica BOND™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins. The section was then incubated with ab33780, 1ug/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset secondary-only control image is taken from an identical assay without primary antibody.

ab33780 staining Glucose Transporter GLUT4 in HeLa cells. The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% PBS-Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at 4°C with ab33780 at 1µg/ml and ab7291, Mouse monoclonal [DM1A] to alpha Tubulin - Loading Control. Cells were then incubated with ab150081, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 488), pre-adsorbed at 1/1000 dilution (shown in green) and ab150120, Goat polyclonal Secondary Antibody to Mouse IgG - H&L (Alexa Fluor® 594), pre-adsorbed at 1/1000 dilution (shown in pseudocolour red). Nuclear DNA was labelled with DAPI (shown in blue).

Image was acquired with a high-content analyser (Operetta CLS, Perkin Elmer) and a maximum intensity projection of confocal sections is shown.

Immunohistochemical of PFA-fixed paraffin-embedded rat skeletal muscle skeletal tissue, labelling glucose transporter GLUT4 with ab33780 at a dilution of 1/200 incubated for 13 hours at 4°C in 1% BSA in TBS. Antigen retrival was with Tris-EDTA at pH 9.0 (heat mediated). Blocking was with 3% BSA incubated for 1 hour at 37°C. Secondary was a Goat anti-rabbit polyclonal Akaline Phosphotase conjugate at 1/200.

Immunofluorescence staining of GLUT4 using ab33780 inab277612, which were differentiated for 3 (left panel), 5 (middle panel) and 10 days (right panel) post induction.

The cells were fixed with 100% MeOH (5 min) and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab33780 at 5 μg/mL and ab7291, Mouse monoclonal [DM1A] to alpha Tubulin, at 1/1000 dilution. Cells were then incubated with ab150081, Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed at 1/1000 dilution (shown in green) and ab150120, Goat Anti-Mouse IgG H&L (Alexa Fluor® 594) preadsorbed at 1/1000 dilution (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).

Glucose transporter GLUT4 also known as GLUT-4 or GLUT 4 transporter plays an important role in glucose uptake in cells. It is an insulin-responsive class I facilitated glucose transporter. GLUT4 predominantly expresses in insulin-sensitive tissues such as adipose tissue and skeletal and cardiac muscles. The functional transport mechanism facilitates glucose entry into the cell by translocating from intracellular vesicles to the plasma membrane in response to insulin stimulation. The molecular weight of GLUT4 is about 50 kDa and this property is often observed in techniques such as GLUT4 western blot where it aids in the identification of the transporter.
Biological function summary

GLUT4 helps regulate blood sugar levels through its control over glucose uptake in muscle and adipose tissues. As an important component of the insulin-regulated glucose transport system GLUT4 responds to increased insulin levels by moving to the cell's surface aiding in glucose transportation into the cell. This adaptive response reduces blood glucose concentration after food intake. GLUT4 operates largely independently but may interact with other glucose transporters like GLUT1 to maintain normal cellular glucose homeostasis.

Pathways

GLUT4 plays a significant role in the insulin signaling pathway and glucose homeostasis. Response to insulin is regulated by its intracellular localization : in the absence of insulin, it is efficiently retained intracellularly within storage compartments in muscle and fat cells. Upon insulin stimulation, translocates from these compartments to the cell surface where it transports glucose from the extracellular milieu into the cell.
