Supplementary information

GLUT5 also known as SLC2A5 is a fructose transporter protein that is part of the class II facilitative glucose transporter family. This protein has a molecular mass of approximately 58 kDa. It is highly expressed in the small intestine where it facilitates the uptake of dietary fructose into enterocytes. GLUT5 is also found in other tissues including the kidney testis skeletal muscle and fat tissue but at lower levels.
Biological function summary

GLUT5 plays a vital role in fructose metabolism and energy homeostasis. It functions as a carrier protein mediating the passive transport of fructose along its concentration gradient. GLUT5 does not form a complex with other transporters but works alongside them like GLUT2 which is responsible for fructose export from enterocytes into the bloodstream.

Pathways

GLUT5 is significantly involved in carbohydrate digestion and absorption pathways. It interacts with proteins like GLUT2 during the process of fructose transport across the intestinal epithelium. This functional interaction is key in maintaining normal blood sugar levels and ensuring efficient nutrient absorption post-digestion.

Improper function of GLUT5 relates to fructose malabsorption and metabolic conditions such as obesity. Fructose malabsorption can cause gastrointestinal distress and contribute to irritable bowel syndrome. Furthermore GLUT5 expression studies in obesity link it to other proteins like leptin which influences appetite and energy balance.

Target data

Functions as a fructose transporter that has only low activity with other monosaccharides (PubMed : 16186102, PubMed : 17710649, PubMed : 28083649, PubMed : 29548810, PubMed : 8333543). Can mediate the uptake of 2-deoxyglucose, but with low efficiency (PubMed : 1695905). Essential for fructose uptake in the small intestine (By similarity). Plays a role in the regulation of salt uptake and blood pressure in response to dietary fructose (By similarity). Required for the development of high blood pressure in response to high dietary fructose intake (By similarity).
