HeLa (human epithelial cell line from cervix adenocarcinoma) cells stained for GMDS (Green) using ab238613 at 1/133 dilution in ICC/IF, followed by Alexa Fluor® 488-congugated Goat Anti-Rabbit IgG (H+L) secondary antibody.

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The nuclear counterstain was DAPI (Blue).

Paraffin-embedded human breast cancer tissue stained for GMDS using ab238613 at 1/400 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

The GMDS protein also known as GDP-mannose 46-dehydratase plays an important role in catalyzing the conversion of GDP-mannose to GDP-4-keto-6-deoxymannose one step in the biosynthesis of GDP-fucose. It has an approximate mass of 39 kDa and predominantly expresses in the liver kidneys and certain immune cells. The GMDS enzyme operates in the cytosol contributing to cellular metabolism and energy balance providing substrates necessary for further enzymatic conversions.
Biological function summary

GMDS contributes significantly to the fucosylation process a modification critical for the functioning of various glycoproteins and glycolipids. It participates in the synthesis of fucose which serves as a substrate for fucosyltransferases in glycan decorations.
