Immunohistochemical analysis of paraffin-embedded human mammary gland tissue labeling GNAQ with ab210004 at 1/1000 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) Ready to use. Immunoprecipitation - Anti-GNAQ antibody [EPR20978] (AB210004)
GNAQ was immunoprecipitated from 0.35 mg of A549 (human lung carcinoma cell line) whole cell lysate with ab210004 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab210004 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10000 dilution.
Lane 1 : A549 whole cell lysate 10 μg (Input).
Lane 2 : ab210004 IP in A549 whole cell lysate.
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab210004 in A549 whole cell lysate.
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 3 minutes.

Lanes 1 - 2 : Merged signal (red and green). Green - ab210004 observed at 42 kDa. Red - loading control, ab18058, observed at 130 kDa.
ab210004 was shown to specifically react with GNAQ in wild-type HAP1 cells as signal was lost in GNAQ knockout cells. Wild-type and GNAQ knockout samples were subjected to SDS-PAGE. ab210004 and ab18058 (Mouse anti-Vinculin loading control) were incubated overnight at 4°C at 1/1000 dilution and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.

Western blot - Anti-GNAQ antibody [EPR20978] (AB210004)
ab210004 was shown to react with GNAQ in wild-type HAP1 cells in Western blot with loss of signal observed in a GNAQ knockout cell line. Wild-type HAP1 and GNAQ knockout cell lysates were subjected to SDS-PAGE. Membranes were blocked in 5% milk in TBST for 1 hr before incubation with ab210004 overnight at 4°C at a 1/1000 dilution. Blots were incubated with secondary antibodies at 0.2 µg/mL before imaging.

Biological function summary
GNAQ influences multiple cell processes mediating the activation of phospholipase C-beta a critical enzyme in hydrolyzing phosphatidylinositol 45-bisphosphate (PIP2) to inositol trisphosphate and diacylglycerol. This action leads to the release of calcium ions from intracellular stores. GNAQ acts as part of a heterotrimeric G protein complex which requires coordination with beta and gamma subunits to achieve full functional capability. Its activity is critical for processes like cell proliferation and differentiation.
Pathways
GNAQ notably participates in the phosphoinositide signaling pathway and calcium signaling pathway. In the phosphoinositide signaling pathway GNAQ facilitates the link between G protein-coupled receptors (GPCRs) and downstream effectors such as phospholipase C-beta. Its activity couples with proteins like GNA11 another member of the Gq family which often shares overlapping functions and roles within these pathways. The calcium signaling pathway involves GNAQ-induced release of calcium ions contributing to various cellular events.
