GNMT also known as Glycine N-methyltransferase functions by catalyzing the methyl transfer from S-adenosylmethionine (SAM) to glycine which results in the formation of N-methylglycine. This enzyme possesses a molecular mass of approximately 292 amino acids. GNMT expresses strongly in the liver but also occurs in other tissues like the pancreas. It maintains the balance of SAM and influences cellular methylation processes.
Biological function summary

GNMT plays a vital role in regulating the cellular methyl group pool. It lacks association with any complex; instead GNMT works independently to control the levels of SAM influencing numerous methylation reactions. Its activity helps in maintaining normal liver function and detoxification processes within the body. Furthermore GNMT impacts the metabolism of several amino acids.

Pathways

GNMT participates in the methionine cycle and folate metabolism pathways. In the methionine cycle GNMT regulates SAM levels alongside proteins such as methionine adenosyltransferase and S-adenosylhomocysteine hydrolase which influence methylation activities broadly. Catalyzes the methylation of glycine by using S-adenosylmethionine (AdoMet) to form N-methylglycine (sarcosine) with the concomitant production of S-adenosylhomocysteine (AdoHcy), a reaction regulated by the binding of 5-methyltetrahydrofolate. Plays an important role in the regulation of methyl group metabolism by regulating the ratio between S-adenosyl-L-methionine and S-adenosyl-L-homocysteine.
