Function

Constitutively active G-protein coupled receptor that maintains high 3'-5'-cyclic adenosine monophosphate (cAMP) levels that a plays a role in serveral processes including meiotic arrest in oocytes or neuronal development via activation of numerous intracellular signaling pathways. Acts as an essential activator of thermogenic adipocytes and drives thermogenesis via its intrinsic G(s)-coupling activity without the requirement of a ligand (PubMed:34048700). Has a potential role in modulating a number of brain functions, including behavioral responses to stress (By similarity), amyloid-beta peptide generation in neurons (By similarity). Stimulates neurite outgrowth in cerebellar granular neurons modulated via PKA, ERK, and most strongly PI3K-mediated signaling pathways (By similarity).

Activity summary

GPR3 also known as G protein-coupled receptor 3 is a member of the GPCR family. This receptor weighs approximately 37 kDa and exists in the cell plasma membrane where it triggers intracellular signaling pathways by interacting with G proteins. Expressed widely in the central nervous system GPR3 plays a mechanical role by activating adenylate cyclase and increasing cyclic AMP (cAMP) levels without any ligand binding a unique characteristic of this receptor.

Biological function summary

The function of GPR3 influences neural processes through its regulation of cAMP signaling. GPR3 is notably a part of signaling cascades within neuronal tissues and impacts processes such as neurogenesis and synaptic transmission. The receptor does not act alone; cooperation with other proteins in cAMP-independent pathways suggests possible involvement in broader complexes affecting brain function.

Pathways

GPR3 contributes significantly to the modulation of signaling via pathways like the cAMP/PKA pathway and may overlap or interact with the Wnt signaling pathway. These pathways include proteins like adenylate cyclase which GPR3 activates playing roles in cellular communication and signal transduction. Through these systems GPR3 impacts various physiological processes especially in the central nervous system intertwining with additional GPCR family members and intracellular signaling proteins.

Associated diseases and disorders

GPR3 reveals links to Alzheimer's disease and depression potentially due to its regulation of cAMP and related impacts on brain function. The receptor's interaction with proteins in these conditions suggests a possible contribution to disease pathology. Specifically its relationship with beta-amyloid proteins in Alzheimer's elucidates a connection of GPR3 to neurodegenerative pathways. Similarly altered GPR3 function and its effects on serotonin pathways provide insights into depression mechanisms and therapies.
