GPR75 also known as G protein-coupled receptor 75 is a member of the large GPCR family characterized by seven transmembrane domains. It is encoded by the GPR75 gene and exhibits a molecular mass of approximately 54 kDa. This receptor is expressed in various tissues including the brain and immune cells suggesting its involvement in multiple physiological roles. As a GPCR GPR75 interacts with G proteins to transmit extracellular signals across the cell membrane initiating intracellular responses.
Biological function summary

GPR75 is implicated in inflammatory responses and neuronal signaling. It is not known to be part of a larger protein complex but functions as a receptor interacting with ligands to modulate various cellular activities. Its expression in immune cells suggests a role in modulating immune responses while in the brain it might influence neural circuitry and communication.

Pathways

GPR75 interacts with important signaling networks. Notable among these are the cAMP-mediated signaling pathway and the calcium signaling pathway. Through these pathways GPR75 participates in processes important for cellular responses to external stimuli. It shares pathway relations with other GPCR family members such as the adenosine receptors which are also known to modulate neurotransmission and inflammatory responses.

Studies have suggested a link between GPR75 and obesity as well as neurodegenerative diseases. Changes in GPR75 function may influence metabolic regulation and energy homeostasis potentially impacting obesity. In neurodegenerative contexts altered signaling by GPR75 might affect neuronal survival and function contributing to disease progression. Its role in these disorders may involve interactions with proteins like insulin receptors in obesity and tau proteins in neurodegenerative conditions.

Target data

G protein-coupled receptor that is activated by the chemokine CCL5/RANTES. Probably coupled to heterotrimeric Gq proteins, it stimulates inositol trisphosphate production and calcium mobilization upon activation. Together with CCL5/RANTES, may play a role in neuron survival through activation of a downstream signaling pathway involving the PI3, Akt and MAP kinases. CCL5/RANTES may also regulate insulin secretion by pancreatic islet cells through activation of this receptor.
